| After the sampling process comes the extraction of the DNA from the bone material. Firstly the sample is reduced to bone powder. The sample material is crushed in a zirconiumoxide covered grinding beaker in a mineralogical mixer mill. This mechanical milling is followed by a decalcification step using EDTA. Thereby osteocytes are released from the hydroxylapatite. At the end of this lysis step, the DNA is settled out and can now be isolated through phenol chloroform extraction and cleansed from the other cell components. Following this, a further cleansing step is taken to separate the DNA from further impurities and the concentration is increased through a filter membrane. The thus obtained DNA extract can now be used in a subsequent PCR (polymerase chain reaction) for the amplification of a particular gene locus. |
